The investigation of the effects of micropatterns and cell-adhesive peptide on myoblast function using cell-interactive hydrogels |
초록 |
It is well known that chemical and structural characteristics of underlying substrate can regulate cell adhesion and growth for tissue regeneration. However, there has been no report on whether the influences of these factors are synergistic or independent to each other. Given that, we prepared peptide-incorporated hydrogels of (1) various peptide density (0.19 ± 0.04 to 0.89 ± 0.08 nmol/cm2) and (2) micro-scaled grooves with various widths (10, 25, 80 μm). We observed the C2C12 skeletal myoblasts and human smooth muscle cells cultured on the hydrogels were sensitive to peptide concentration or surface geometry, which regulate cellular adhesive morphology, proliferation, and distinct distribution of focal adhesion proteins. Our newly developed cell-adhesive peptide-incorporated hydrogels with defined patterns may be effective in investigation of the effect of these factors on cellular responses and elucidation of regulatory mechanisms underlying cell-ECM communication. |
