Tannic acid, a galloylated glucose, can bind reversibly to various polymers and biopolymers, such as protein and nucleic acids, via hydrogen bond and hydrophobic interaction. Herein we propose a simple, versatile, and efficient protein loading strategy into premade hydrogels using tannic acid. After immobilizing tannic acid on various premade hydrogels by simple incubation, green fluorescent protein as a model protein drug was loaded into tannic acid-coated hydrogels. Tannic acid-coated hydrogels showed higher protein loading efficiency and more sustained release of protein compared to bare hydrogels. The release rate can be controlled by the concentration of tannic acid. This novel platform for protein loading into premade hydrogels can be applied for delivery of protein therapeutics and development of cell culture matrix. |
