The effect of porosity according to different amount of HPC on drug release from multi-coated bead |

Among various drug delivery systems available in recent pharmaceutical industry, Oral delivery systems hold the major part because of their evident advantages of easy administration and superior patient compliance. So we purposed to prepare new device of granules containing water-insoluble drug. We prepared osmotic multi-coated granules by fluidized-bed spray coating method, and model-drug used nifedipine(ND). To improve release profile of ND beads, water absorption force, bead size and membrane porosity are very important. So obtained beads were coated with membrane containing different amount of hydroxypropyl cellulose(HPC) to form channels water passed. Porosity of membrane could be controlled by water soluble excipient such as HPC to control drug release. As increasing amount of HPC, channels passing inner part of multi-layered granule are increased. Consequently, drug release increased as amount of HPC increased.
