Hyaluronic acid and PEG-TRAIL ionic complex as a promising therapeutics for rheumatoid arthritis |

In this study we developed nanocomplex formulations which can stabilize PEGylated-TRAIL (PEG-TRAIL) and exert the therapeutic effect for rheumatoid arthritis. The positively charged (PEG-TRAIL) was mixed with negatively charged hyaluronic acid (HA), resulting in the formation of nanocomplex (PEG-TRAIL/HA). To evaluate the in vivo biodistribution and release kinetics, mice were given Cy5.5-labeled PEG-TRAIL by subcutaneous injection of three different formulations and monitored as a function of time using near-infrared optical imaging system. These results imply PEG-TRAIL was released in a sustained manner from the nanocomplex in 1% HA solution. In a collagen induced arthritis mouse model, the therapeutic effect was significantly enhanced as determined by the clinical scores and histology. The production of RA specific serum inflammatory cytokines and collagen specific antibodies were significantly reduced due to the sustained delivery of PEG-TRAIL. |
