Low molecular weight poly(ethylene imine)-methyl cellulose conjugate for gene delivery systems |

Methyl cellulose(MC) has been extensively used for biomedical use. As a gene vector, we synthesized methyl cellulose-g-poly(ethylene imine)(MC-PEI) through grafting low molecular weight(0.8k) branched poly(ethylene imine) to periodate-oxidized methyl cellulose by reductive amination. MC-PEI was able to condense plasmid DNA(pDNA) efficiently and protect pDNA from serum attack. The particle sizes of MC-PEI/pDNA polyplexes are below 160 nm, and the zeta potentials were around 35 mV. The MC-PEI/pDNA polyplexes had well-formed spherical shapes. In vitro cell viability and transfection efficiency were evaluated in C2C12 and Hela cells. The cytotoxicities of MC-PEI was much lower than that of PEI25k and even that of PEI 0.8k in both cell lines. The gene transfection efficiency of MC-PEI polyplexes at w/w ratio of 75 in both cell lines was comparable to that of PEI25k polyplexes at w/w ratio of 1. These results suggest potential application of MC-PEI for non-viral gene delivery system. |
