Chitosan-graft-guanidinylated polyethyleneimine (CS-PEI-G) was synthesized for gene delivery systems. The conjugation degree of PEI was 11.7% and the molecular weight of CS-PEI-G was measured to be 164.2 kDa. CS-PEI-G was able to retard pDNA at a weight ratio of 1, and formed stable polyplex with pDNA. Because of guanidine moieties which may make cell membrane destabilized, CS-PEI-G showed a moderate cytotoxicity at a high polymer concentration, but it showed improved transfection efficiency in all weight ratios compared to CS-PEI regardless of serum. Especially in the presence of serum, transfection efficiency of CS-derivatives represent small decrease. This indicates that serum stabilities of CS-derivatives are better than that of PEI25k. According to flow cytometry results, improved transfection efficiency of CS-PEI-G may be attributed to the nuclear localization ability of the guanidine moieties. This study has demonstrated that CS-PEI-G has a potential for gene delivery carriers. |
