In this study, we synthesized cross-linked poly(propylene imine) (PPI) dendrimers having bio-reducible linkages (PPI-CBAs) for gene delivery systems by using N, N’-cystamine bisacrylamide (CBA) as a cross-linker which has a disulfide bond. The polymers could retard DNA at low weight ratios, meaning good DNA condensing ability. Most of Zeta-potential values of the polyplexes were located between 20 mV and 60 mV and the size of polyplexes was relatively consistent and almost lay between 100 nm and 200 nm, suggesting the polyplexes were suitable for cellular uptake. The cytotoxicity of the polymers were relatively high and increased along with CBA ratios. The transfection efficiency of the polymers were lower than PEI25k in serum free condition. However, the efficiency became comparable to that of PEI25k in the presence of serum. From these results, it can be thought that PPI-CBAs have a potential for gene delivery systems and cytotoxicity issues should be solved in further study. |
