제목 Quantifying cell-material interactions using FRET technique
초록 Alginate was chemically conjugated with adhesion ligands (e.g., G4RGDASSK), and RGD island spacing in the gel was varied from 36 to 78 nm while the average bulk ligand density was kept constant. We found that the mechanical properties of the gels were not altered by introducing ligands in the gels. The strength of cell adhesion was quantified using fluorescence resonance energy transfer (FRET) technique and cell adhesion index (CAI). FRET is a technique that can quantify the amount of energy transfer from donor to acceptor molecule. CAI was used to assess the relative number of cells remaining on the gel after application of centrifugal forces. Both methods have shown the same trend related to cellular adhesion onto the ligand-organized gels. These results support that biophysical cues such as ligand organizations on synthetic extracellular matrices play an important role to modulate cellular responses to its microenvironment.
