Enzymatic in situ formation of hydrogels from gelatin-tyramine and chitosan 4-hydroxylphenylacetamide for delivery of human adipose stem cell and vasculogenic induction |

Injectable hydrogels based on proteins are highly preferred as they can favorably interact with the encapsulated cells via specific peptide epitopes and also undergo cell-mediated enzymatic degradation. Therefore, extracellular matrix (ECM)–derived proteins, such as collagen, gelatin, laminin, and elastins are potential candidates to develop cell delivery vehicles. Hydrogels consisted of the tyramine-conjugated gelatin (GTA) and chitosan-4-hydroxylphenylacemide (CHPA) was obtained by enzymatic crosslinking under physiological conditions using horseradish peroxidase (HRP) as a catalyst and hydrogen peroxide (H2O2) as an oxidant. The gel formation was varied within a time period of 20 to 180 seconds by controlling the concentration of polymers and HRP. The physical properties and the degradation of hydrogel were confirmed by degradation test. Human adipose stem cells are encapsulated in the CHPA-GTA in the presence of PDGF for the application of vasculogenic induction. These injectable hydrogel could be suitable for cell, drug and growth factor delivery systems. |
