제목 The use of alginate-hyaluronic acid microbeads as a promising method for cell or drug delivery for the purpose of kidney regeneration
초록 Delivery of cells and growth factors from a hydrogel carrier can be used as a promising method to accelerate the regeneration of injured tissue or organs. In this study, 50-300µm alginate-hyaluronic acid (ALG-HA) injectable microbeads, with the purpose of delivering stem cells for tissue engineering, were prepared by a spraying method into a CaCl2 solution that shows high porosity for the exchange of nutrition and waste. In addition, the size distribution and surface morphology was investigated using optical and scanning electron microscopy, respectively. The chemical structural properties of the ALG-HA microbeads were examined by X-ray diffraction and Fourier transforms infrared spectroscopy. Their biocompatibility was examined using both in vitro and in vivo studies. For this purpose, rat bone marrow stem cells were encapsulated into microbeads to investigate the cell release, cell viability, and proliferation behaviour up to 21 days, and cells were found to be released from the microbeads after 7 days. Furthermore, the in vivo biocompatibility of ALG-HA microbeads was examined using microbeads without cell encapsulation under the kidney capsule, in order to assess the foreign body reaction and the inflammatory response for 14 days. The desired in vivo response to hydrogel makes them exquisite candidates for subcapsular cell and drug delivery to kidney tissue.
