Injectable in situ forming chitosan/gelatin based hydrogel incorporating with bi-calcium phosphate nanoparticle and BMP-2 as filler for prevention of bone fracture |

The goal of this study is the development of a novel injectable system which is in situ forming biomimetic bone filler consisted of the modified gelatin and chitosan hydrogel incorporating with bi-calcium phosphate (BCP) and bone morphogenetic protein-2 (BMP-2). The system mainly consists of tyramine-conjugated gelatin (GTA) and chitosan-4-hydroxylphenylacemide (CHPA), which can be formed to gel phase via enzymatic reaction using horseradish peroxidase (HRP) and hydrogen peroxide (H2O2) under physiological conditions. To improve the capabilities of osteogenesis, BCP nanoparticles and BMP-2 were loaded into this system. The CHPA-GTA with BCP/BMP-2 hydrogel was characterized by scanning electron microscopy, electron dispersion spectroscopy, and X-ray diffraction. The hydrogel structure showed a porous interconnected morphology. Biocompatibility of this hydrogel was evaluated using MC3T3-E1 via MTT assay and F-actin staining. The hydrogel was maintained in viscous phase after injection with cells, showed suitable degradation rate and improved cell viability and proliferation. These results highlight the potential of using a novel injectable biomimetic scaffold for prevention of bone disease. |
