제목 | pH sensitive imidazole grafted poly(amino acid) nanoparticle |
초록 | Polymers that respond to a small change in pH may find a wide range of applications. In particular, much attention has been paid in the last two decades to pH-sensitive polymers for drug delivery application. In case of biological and pharmaceutical systems which need small fluctuation in pH around 7 such as intracellular endosomal drug release and tumor targeting delivery, pH sensitive polymers are very important. And amino acid-based polymers including polypeptides have been developed due to their wide potential applications for pharmaceutical and biomedical applications. The protein like amide linkages in poly (amino acid) offer biodegradable properties to polymer. Recently self-assemblies of polymer amphiphiles have been studied. Not only block copolymers but also graft copolymers of amino acid have attracted attention, because they can be easily designed and synthesized. Poly(succinimide) (PSI) was easily reacted with a primary amine to produce biodegradable poly(amino acid) such as poly (asparagine) and poly(aspartic acid)4. Our study aims to prepare pH-sensitive amphiphilic poly(amino acid) nanoparticle by grafting sulfadimethoxine (pKa=6.2) and histamine to PSI for intracellular drug delivery system. The polymers had pH response abilities according to small pH change (pH 7→5) and aggregated in aqueous solution by hydrophobic interaction. Turbidity test, buffering effect test, and dynamic light scattering are performed for determining the pH sensitive properties and the size distribution of nanoparticle. |
