Prostate cancer is one of the major mortality disease and the research on its treatment has been active. There are several studies using siRNAs that inhibit the activity of the androgen receptor which is specifically present in prostate and helps growing of prostate, but more effective treatments are still required. In this study, we propose to treat prostate cancer more effectively by delivering both several kind of siRNAs and anticancer drug at once. We developed a colloidal formulation (< 10 nm) of the hydrophobically associated multiple monocomplex (HMPlex) which consists of siRNAs, apoptotic-anticancer drug, and Pluronic F-68 to deliver to prostate cancer cells. We have compared various combinations of siRNAs in the HMPlex in vitro and in vivo, and the results showed that simultaneously silencing various genes was more effective in inducing cell apoptosis than silencing a single gene in prostate cancer cells. |
