제목 Self-Assembled Polymer Nanoparticles for Molecular Imaging
초록 Polymer amphiphiles form polymeric micelles or self-assembled nanoparticles by the interactions between the hydrophobic segments in aqueous environment. Both the polymeric micelle and nanoparticle share common characteristics of a unique core-shell structure with narrow size distribution. The inner core serves as a nano-container to deliver various chemicals and biological molecules used for diagnosis or treatment of certain diseases. A prolonged circulation time in blood stream is owing to the hydrophilic outer shells by avoiding the reticuloendothelial system (RES) and is known as essential requirements to target specific cells, tissues, and organs in our body. Disintegration of the assembled structure, which results in the release of contents in the inner core, can be controlled by the activity of specific enzymes in target sites.
The self-assembled nanoparticles, activatable for caspase-3, are designed and characterized. A NIR cyanine fluorochrome was served as an optical reporter to image the activity of the protease. Results of in cell challenge experiments will be presented and discussed in this study.
