제목 Controlled Bioconjugation of Poly(poly(ethylene glycol)methyl ether methacrylate)with Green-Fluorescent Proteins
초록 We developed a new approach to bio-conjugate proteins and polymers. The developed method is based on the strong interaction with a sequence of histidine residues (His-tag) in genetically engineered proteins and Ni2+ complexed nitrilotriacetic acid (NTA) group at the polymer chain end. In our study, green fluorescent protein (GFP) with His-tag was conjugated with Ni2+-NTA end-functionalized poly(ethylene glycol) methyl ether methacrylate. His-tagged GFP was expressed in recombinant E. coli BL21 (DE3) and end-functionalized poly(ethylene glycol)methyl ether methacrylate was prepared by Atom Transfer Radical Polymerization using NTA containing initiator. As His-tagged proteins is well-known to have specific binding affinity with Ni2+-NTA, His-tagged GFP was able to be conjugated with Ni2+-NTA end-functionalized poly(ethylene glycol)methyl ether methacrylate in a controlled fashion.
