제목 중간엽 줄기세포와 고분자 복합체를 이용한 연골 재생의 최근 동향
초록 Autologous chondrocytes transplantation (ACT) was developed for the treatment of cartilage defects. However, some problems such as the uneven distribution of grafted chondrocytes and changes of chondrocytes phenotype in vitro have been reported. However, mesenchymal stem cells (MSC) can be successfully differentiated into chondrocytes under defined culture conditions. We investigated the biological and mechanical properties of regenerated cartilage using MSCs and poly lactic-co-glycolic acid (PLGA) scaffold pretreated of TGF-β3. The composites were transplanted to the osteochondral defect in rabbit knee and 10 of the 12 rabbits showed cartilaginous regeneration after 12 weeks post-transplantation. In indentation test, this had ca. 80% of Young’s modulus of normal articular cartilage. Histological data demonstrated hyaline like cartilage with glycosaminoglycan and type II collagen, suggesting that this treatment could be possible as another concept of cartilage tissue engineering.
