Fabrication of Hydroxyapatite(HAp) granules loaded Oxidized Alginate-Gelatin-Biphasic Calcium Phosphate(Oxg Alg-Gel-BCP) hydrogel complex and its in-vitro evaluation for bone tissue regeneration |

There are many reports on the fabrication of injectable bone substitutes which are combined with bio-ceramics and bio-polymer for bone tissue engineering. In this work, we fabricated spherical HAp granules and loaded into Oxidized Alginate-Gelatin-Biphasic Calcium Phosphate in order to use as a bone substitute system. For the fabrication of HAp granules, PVA-Gel-HAp composite slurry was dropped first by a syringe through the nozzle on liquid nitrogen maintaining the flow rate, distance of dropping and then lyophilized for 2 days. Then polymers were burnt out from HAp granules by tube furnace and finally sintering occurred at 13500 C. These HAp granules were loaded into Oxg Alg-Gel-BCP slurry and freeze-dried for 2 days to obtain porous structure. Detailed morphological and material characterizations such as microstructure analysis, compressive strength, porosity, FTIR spectra were carried out. MTT assay, confocal imaging and live-dead assay were carried out as in vitro experiments to observe the cell attachment, proliferation with Rat Bone Marrow Mesenchymal Stem Cell (RBMSC). |
