Development of a BCP-chitosan based injectable bone substitute for tissue engineering |

The aim of this study is to develop an injectable bone substitute (IBS) using polymer and ceramic based granules. The porous BCP blocks were fabricated by sponge replica method, crushed and sieved to make granules, resulting in particles < 1mm in size. These particles were mixed with a phenol conjugated chitosan solution and freeze-dried for 24hr. These granules coated with chitosan were loaded in syringe and cross-linked by adding HRP/H2O2 solution in PBS before use. The IBS was characterized by scanning electron microscopy (SEM), electron dispersion spectroscopy (EDS), X-ray diffraction (XRD). The SEM result showed that the BCP granules were connected within chitosan hydrogel construct. The BCP granules loaded IBS exhibited excellent biocompatibility, cell proliferation and cell adhesion behavior resulted in the MTT assay and immunofluorescence analysis using bone marrow messenchymal stem cells (BMSCs). Based on these results, this IBS could be considered as potential cell delivery system and bone tissue engineering applications. |
