제목 | In vitro Osteogenesis of Bone Marrow Stromal cells on SIS/PLGA Scaffold |
초록 | Bone marrow stromal cells (BMSCs) have pluripotent differentiation potential including differentiation into osteoblasts, chondrocytes, adipocytes, and muscle cells.1 Meanwhile small intestimal submucosa (SIS) is a naturally occurring tissue matrix composed of extracellular matrix proteins and various growth factor. SIS is easily derived from the porcine jejunum. Therefore, SIS have the potential in the biomedical application as the scaffold. In this work, we examined the differentiation of rat bone marrow stromal cells (rBMSC) into osteoblasts on SIS/PLGA scaffold in the presence of osteogenic media. rBMSCs were supplemented with DMEM, 10% FBS and 1% penicillin-streptomycin cultured for up to passage 8. Then the rBMSCs were cultured on SIS/PLGA scaffold. The ratio of SIS used was 0, 10, 20 and 50% for weight of PLGA (90,000 g/mole). For in vitro osteogenesis, rBMSCs were supplemented with osteogenic medium. The osteogenic medium consisted of DMEM, 10mM β-glycerophsphate, 50 ㎍/㎖ L-ascorbic acid, 10-7 M dexamethasone, 10-8 M vitamin D3, 10% FBS and 1% penicillin-streptomycin. While, control medium consisted of DMEM, 10% FBS and 1% penicillin-streptomycin. rBMSCs on scaffold maintained for 7, 14, 21 and 28 days. Osteogenic differentiation in vitro was evaluated by means of MTT, ALP activity, PCR, and immunohistochemistry. Cell viability on scaffold increase as culture day incresed and in addition, ALP activity increased. The result of these experiments indicated that rBMSCs differentiated into osteoblasts.
