Method Development for Direct Detection of Glycoproteins on the Aminophenylboronic Acid-Functionalized Self-Assembled Monolayers by Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry |

Glycosylation is one of the most abundant post-translational modifications of proteins and primarily associated with membrane-bound and secreted proteins. Glycoproteins fulfill a wide range of functions in the cell, and new functions are constantly being discovered. Therefore, the discovery and identification of glycosylated peptides and proteins, and the analysis of their glyco-structures are increasingly important in diagnosis and proteomics. This study describes a simple method for direct detection of glycoproteins after affinity-capturing on the plate using a combination of self-assembled monolayers(SAMs) and matrix-assisted laser desorption/ionization mass spectrometry(MALDI). The terminal hydroxyl groups of the SAMs were activated by N,N-disuccinimidyl carbonate, followed by conjugation with aminophenylboronic acid(APB). The formed APB-functionalized SAMs for MALDI plates were successfully applied to the affinity-capturing of a glycoprotein from the protein mixture. |
