Controlled release of Pioglitazone form PLGA nanospheres |

In this work we encapsulated hydrophobic model drug pioglitazone by the PLGA using emulsion-evaporation method. pioglitazone loaded PLGA nanospheres were characterized by SEM, DLS and HPLC measurements. In vitro, the release test for the conformance as controlled drug and MTT assay for toxicity test was performed and the effect of the free pioglitazone and pioglitazone loaded PLGA nanospheres was compared through in vivo. These particulate carriers exhibited controlled in vitro release of pioglitazone for 40days at a nearly constant rate. Pioglitazone loaded PLGA nanospheres confirmed through the MTT assay and animal experiment that toxicity was less than the free drug. In conclusion, these results indicate that pioglitazone loaded PLGA nanospheres have greater potential for effect treatment of diabetes as controlled drug release. |
