Nano structured-fibrous scaffold was fabricated by electrospinning of poly(ε-caprolactone) blended with poly(ε-caprolactone)-poly(ethyleneglycol)[PCL-PEG-NH2] diblock copolymers. Upon increasing blend ratios of the diblock copolymers upto 20 %, average diameters of nanofibers significantly increased, showing PEG chains conjugated to PCL interfered an electrospinning process of PCL nanofibers. Murine fibroblasts were cultivated on the PCL/PCL-PEG nanofibers, confirming PEG had a profound effect on the growth of the cells. In order to facilitate wound healing process, human epidermal growth factor (EGF) was chemically-conjugated on the surface of electrospun nanofibers. For wound healing animal models, C57Bl/6 mice were adminstered with streptozotocin for diabetic symptoms. The electrospun nanofibers with conjugated EGF accelerated wound reparing processes compared to the nanofiber mixed with EGF solution, PCL nanofiber, or control. |
