Low molecular weight water soluble chitosan for accelerating DNA strand exchange |

The association, dissociation and strand exchange of nucleic acids are essential processes in maintenance of living system. DNA strand exchange between double strand and its complementary single strand has a pivotal role in repair of damaged DNA and genetic recombination. In the previous study, we reported that the cationic comb-type copolymer, PLL-g-Dex showed higher efficacy on the accelerating DNA strand exchange. In this study, we investigated the accelerating effect of low molecular weight water soluble chitosan(LMWSC) on the DNA strand exchange reaction between a 20 bp duplex DNA and its complementary single strand. LMWSC has a high content of free amino groups in the chitosan chain, implying increased affinity with DNA. DNA strand exchange was studied by polyacrylamide gel electrophoresis(PAGE) using the duplex strand labeled with fluorescein isothiocyanate(FITC) and its complementary single strand. |
