Cloud point (CP) measurements have been performed to investigate the influence of alcohols (C2-C10OH) and surfactants on the micellar behavior of amphiphilic drug amitriptyline (AMT). In case of shorter-chain alcohols (C4OH or C5OH), the CP increased with increasing alcohol concentration in the presence of a small amount of sodium phosphate (SP) because they partition very little in AMT micelles. On the other hand, it steeply decreased with increasing alcohol concentration in case of longer-chain alcohols (C8OH or C10OH) due to their considerable partitioning in AMT micelles. The CP increases on addition of ionic surfactants (alkyltrimethylammonium halide and sodium alkyl sulfate) in the presence of small amounts of SP. Furthermore, the CP increases with the alkyl chain length of the ionic surfactants. The CP becomes less sensitive to addition of ionic surfactants in the presence of large amounts of SP. Addition of Pluronic F-127 to the AMT solution increases the CP, whereas addition of Pluronic 10R-8 decreases the CP. |
