Purification and Identification of Cancer Markers Using Temperature Controllable Bead Affinity Chromatography Chip Based on MALDI-TOF Analysis |

Mico-bead affinity chromatography (μ-BAC) has been successfully utilized in purifying and concentrating biologically important proteins in a microchip system. We have fabricated the temperature controllable μ-BAC chip equipped with two separated chambers which contained aptamer-immobilized affinity beads and trypsin-immobilized beads for purifying and identifying the marker proteins. The marker proteins (HCV and AFP) were captured and purified in the first chamber and transferred to the second chamber after thermal releasing, and digested by the immobilized trypsin. Finally, the digested peptides were analyzed and identified by MALDI-TOF. |
