Thrombin Activatable CT/Optical Dual Imaging Probe Based on Gold Nanoparticles |

AuNPs have attracted scientist’s attention that developed AuNPs as an imaging agent. In this study, we developed thrombin activatable CT/NIRF imaging probe which can detect thrombosis in the blood stream. The particles were coated with silica layer to have stability and biocompatibility in human body. Then thrombin activatable peptide conjugated with dye was conjugated to the nanoparticle surface. For its high X-ray absorption, AuNP can be used as CT contrast agent and NIRF dye for optical imaging. At normal state, nanoparticle conjugated with NIRF dye is quenched because of the NSET between AuNP and dye. However, when the probe is near thrombin, the reaction by thrombin cleaves the peptide and dequenches NIRF dye which makes detecting thrombosis possible. These feasibilities of CT/NIRF dual imaging probe were showed promising results for detecting thrombosis. |
