Applications of self-assembled PEGylated albumin nanoparticle in cancer chemotherapy and theranostics |

Paclitaxel (PTX) is used as a major antitumor agent for the treatment of recurrent and metastatic breast cancer. The clinical application of PTX requires oil/detergent-based solvent due to its poor solubility in an aqueous medium. However, the formulation causes hypersensitivity reactions and limited tumor distribution. Herein, we introduce an oil-free method to prepare albumin-based PTX nanoparticles for systemic cancer therapy using a pegylated human serum albumin (HSA). Simple film casting and rehydration procedure produced PTX-incorporated self-assembled HSA-PEG nanoparticles. The HSA-PEG nanoparticle mediates efficient cellular delivery with the highest cytotoxicity in various breast cancer cell lines. When systemically administered in a mouse xenograft model for human breast cancer, the HSA-PEG nanoparticle formulation exhibited a long systemic circulation and enhanced intratumoral accumulation, resulting in a remarkable anticancer effect and prolonged survival of the animals. |
