제목 Mesoporous silica nanoparticles to induce STING activation for enhanced cancer immunotherapy
초록 Stimulator of interferon genes (STING) pathway activation by cyclic dinucleotide (CDN) has been recently focused for treatment of cancer. STING receptor is one of types pattern recognition receptor (PRR) located in the cytoplasm. Upon an interaction with cGAMP, one type of CDNs, STING receptor secretes Type 1 IFN-B that activate antitumor immune response through maturation of dendritic cells and proliferation of cytotoxic T-cell. However, biodistribution of CDNs to target site is limited due to its enzymatic degradation and hydrophilic nature that makes endocytosis difficult. In addition, most of previous studies carrying CDNs in the nanoparticles have been focused on intratumoral injection to evoke anti-tumor response in tumor microenvironment. In this research, we developed mesoporous silica nanoparticles (MSN) as a carrier for CDN and cancer-specific antigen to induce a strong cancer antigen-specific immune response by delivering CDNs that activates STING receptors and directly delivering of antigen to dendritic cell for antigen presentation in the lymph node. Thus, MSN with CDN and antigen would promote antigen specific cytotoxic T cell for treatment of cancer diseases.
