Nano-particle systems usually present some degree of tumor targeting based on the EPR effect. Previously, we reported that Chitosan-functionalized, Pluronic-based nano-carriers also showed a good tumor targeting. However, like all other kinds of targeted nano-systems, they showed fluorescence signals from non-specific distribution of the nano-carriers, especially from liver. To overcome these background signals, we prepared the hydrocyanine-conjugated nano-carriers (Hydrocyanine-NC), thus they can detect reactive oxygen species (ROS) in tumor pathological site. The hydrocyanine labeling of nano-carriers resulted in almost complete disappearance of fluorescence signal from liver, and it produced strong fluorescence emission to monitor the target tumor site selectivity.
