제목 Development of an in vitro clostridioides difficile infection model for the investigation of host-anaerobe pathogen interactions via integrative multiomics approaches
초록 Clostridioides difficile is an enteric anaerobe pathogen and produces several virulence factors. Although the severity of C. difficile on the host is well known, the exact pathogenic mechanisms and biological pathways of C. difficile infection (CDI) are not yet clear. Here, we developed an in vitro CDI model to investigate host-C. difficile interactions throughout the early (12hr) andlater (48hr) stages of CDI. Intracellular proteomic results confirmed damage inprotein processing and mitochondrial function of Caco-2 cells due to cellular stress caused by CDI. In addition, metabolic results confirmed that several metabolites corresponding to glycolysis, TCA cycle, and amino acid metabolism involved in energy metabolism were decreased. Consequently, our in vitro CDI model and multiomics approach not only provide a better understanding of host-C. difficile interactions but also aid in the treatment of C. difficileand other anaerobic infections.
