Structural studies of collagen and mineral in red sea bream scale |

In this work, we explored the structure and orientation of collagen fibrils and the form of Ca mineral in the scale of a red sea bream, Pagrus major. The synchrotron based small angle X-ray scattering (SAXS) and X-ray absorption spectroscopy (XAS) techniques were employed in this study. The measurements were made at positions along the length of the fish scale, from the younger to the older parts. The SAXS results show that, at many positions, the collagen fibrils are aligned in two directions orthogonal to each other as in the plywood structure. Towards the center of the scale, however, the alignment becomes more random which suggests a stacking disorder during the aging process. The XAS results confirm that Ca in the scale is in a poorly crystalline hydroxyapatite form. Although there is no indication of increased crystallinity during the aging process, a change in the local environment of Ca was observed and could be related to the change in the Ca-O coordination shells. |
