제목 | Development and characterization of a fully human antibody targeting EP4 signaling |
초록 | Prostaglandin E2 (PGE2) is the predominant prostaglandin in solid tumors and promotes tumor growth and metastasis by activating on its receptor, EP1-4. Among them, EP4 promotes migration and invasion in breast cancer. In this study, we developed fully human EP4 antibodies by phage display technique and selected the antibody (Ab5) with the highest binding affinity and specificity through in vitro assays such as enzyme-linked immunosorbent assay (ELISA) and flow cytometry (FACS). Moreover, we found that Ab5 inhibits PGE2-induced migration and invasion in breast cancer cell line, MDA-MB-231, by downregulating signaling molecules related to metastasis. Taken together, our results suggest that Ab5 can be applied as a new therapeutic agent for the treatment of patients with metastatic breast cancer. |
