Optimization of High Cell Density Cultivation and FDH Expression Condition in Methylorubrum extorquens ATCC 55366 by Using Methanol as a Sole Carbon Source |

Carbon dioxide (CO2) is considered one of the main green-house gases. In this study, we have investigated optimized conditions for expression of formate dehydrogenase (FDH), which converts CO2 to formate, at high cell density in Methylorubrum extorquens ATCC 55366 by using methanol as a sole carbon source. At first, we investigated the inhibitory effect of methanol on cell growth in M. extorquens ATCC 55366. Since it was confirmed that cell growth was significantly inhibited in a medium containing upper 3% (v/v) of methanol, we have conducted cultivation to maintain less than 1% of methanol in a medium by controlling the specific growth rate (μ= 0.1 to 0.02). Finally, optical density value (OD600nm) reached over 192 (3.25-fold higher than that of succinate) in 88 hours by using methanol. However, it was confirmed that activity of FDH was significantly low when cells were cultivated in methanol containing medium than that of succinate. To overcome this problem, it was needed to investigate the factors to affect activity of FDH according to carbon assimilation characteristics. |
