Cell-based assays are increasingly being used in drug discovery, genetic analysis and single-cell analysis. Recently significant efforts have focused on the fabrication of cell microarray using a variety of cell patterning techniques such as photolithography or soft lithography. Cell microarrays can be classified into two general categories: chip-based planar arrays and particle-based suspension arrays. Although chip-based planar arrays have been widely used, suspension arrays are becoming popular as an alternative approach due to several advantages over planar array. In this study, we prepared suspension microarrays of cells using various shape of SU-8 microparticles and demonstrate the potential use of this system for multiplexed cell-based assay. Capability of controlling the shapes of microparticles opened the possibility of producing shape-coded suspension array, which can offer a cost-effective and versatile platform for high-throughput and multiplexed cell-based assay. |
