제목 Human DNA purification at high-throughput manner using a centrifugal microfluidic disc for rapid forensic sex-typing
초록 We develop a centrifugal disc for high-throughput human nucleic acid (NA) purification to perform the human sex-typing of forensic samples. As a NA extraction matrix, Whatman glass filter paper was used, and sophisticated passive valves were installed to prevent the buffers from overflowing into the neighboring chamber during the injection. We created a portable workstation that includes a buffer storage system, a buffer injection system, and a rotating device to run the disk. The in-house portable workstation managed the whole operation automatically. The extraction of genomic DNA from thirty forensic samples took just ten minutes. The human alphoid repeat sequence of the Y-chromosome and the human 18sRNA were targeted using purified genomic DNA in a loop-mediated isothermal amplification (LAMP) reaction for sex-typing. We were able to complete the genetic sex-typing in 30 minutes using a high-throughput centrifugal disc for NA extraction and the LAMP reaction.
