MSCs have been widely employed for accelerating tissue regeneration due to its self-renewing, immunomodulatory, as well as inducing angiogenesis. 3D spheroidal model augments the therapeutic capacity of MSCs by inducing hypoxia environment. Thus, we introduced 3D MSCs as a cell-based therapy for treating mucosal ulcer. The 3D MSCs showed a higher gene expression of VEGF, IL-10, and MMP family, which relate to accelerate tissue regeneration, than monolayer MSCs. Thermo-sensitive hydrogel was employed to transfer 3D MSCs on an acid-induced mucosal ulcer of a rabbit. Mucosal ulcer treated with 3D MSCs completely covered with a thicker regenerated mucosal layer compared that with monolayer MSCs at day 7. Also, the regenerated mucosal layer showed a higher expression of keratin 5 and 13 as markers for proliferating of progenitor cells and phenotype of mucosa layer, respectively. Thus, 3D MSCs was confirmed a superior therapeutic efficacy for accelerating regeneration of mucosal ulcer. |
