Hypoxia-responsive Polymeric Nanoparticles Based on Carboxymethyl-dextran for Cancer Therapy |

Hypoxia is a hallmark of various intractable diseases. Herein, we have developed self-assembled nanoparticles which can selectively release the hydrophobic drug in response to hypoxic conditions. To prepare hypoxia-responsive amphiphilic nanoparticles (HR-NPs), hydrophobically modified 2-nitroimidazole (NI) derivative was conjugated to the backbone of carboxymethyl dextran (CM-Dex). Doxorubicin(DOX) as an anticancer drug was encapsulated into HR-NPs. The drug release behavior of DOX from HR-NPs shows significantly high release rate under hypoxia condition, whereas the HR-NPs released DOX in a sustained manner under the normoxic condition (physiological condition). Based on in vitro cytotoxicity tests, HR-NPs show higher toxicity to hypoxic cells than to normoxic cells. From the in vivo biodistribution and therapeutic efficacy tests, it is demonstrated that DOX-loaded HR-NPs have the capability for effective delivery of drugs into hypoxic cells as well as high anti-tumor activity. |
