제목 The combination of high-intensity focused ultrasound and thermosensitive liposomes for improved antitumor efficacy
초록 The combinatory approach of thermosensitive liposomes (TSLs) and high intensity focused ultrasound (HIFU) is emerging as a way of facilitating targeted antitumor drug delivery. Over the last decade, HIFU has been used to induce mild hyperthermia immediately after the systemic administration of doxorubicin (DOX)-loaded TSLs (DOX-TSLs). However, the immediate heating approach (IHA) has limitations such as poor penetration of DOX into the tumor tissue. In this study, a delayed heating approach (DHA) has been explored to maximize the DOX accumulation at tumor. Target tumoral region was pre-treated with HIFU to increase vascular permeability, and then DOX-TSLs were intravenously injected to tumor-bearing mice. DOX-TSLs were selectively accumulated at tumor and the DOX amount was maximized at 6h post-injection. In contrast to IHA, DHA significantly enhanced the antitumor efficacy of DOX-TSLs through the effective penetration to tumor tissue by enhanced permeation and retention effect.
