Tumor-Homing Polymeric siRNA Nanocomplexes(PNCs) for Systemic Delivery in Cancer Therapy |
초록 |
siRNA has been received a great attention as emerging gene therapeutics due to its superior capacity of specific gene silencing on post-transcription level. In our previous studies, rolling circle transcription technique can provide macro molecular weight siRNA as well as a structural formation of siRNA itself. Here, we developed the periodically repeated siRNA through RCT process band its delivery system using glycol chitosan derivatives. The synthesized polymeric siRNA Nanocomplexes(PNCs) has stable formation and protect the siRNA therapeutics against polyanion and nucleases attack formation in blood circulation. For tumor-targeted delivery of siRNA, the EPR effect could be adapted as a passive targeting strategy; immature blood vessel formation and lack of lymphatic drainage in tumor tissues might cause tumor-specific accumulation due to increased permeability and retention of PNC-VEGF in tumor interstitium. |
