Productgion of ACE2-derived peptides in E. coli for the inhibition of spike glycoprotein of SARS-CoV-2 |
초록 |
Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) is highly transmissible and pathogenic coronavirus that emerged in late 2019 and is closely related to severe acute respiratory syndrome coronavirus (SARS-CoV) occurred in 2003. SARS-CoV-2 has spike (S) glycoproteins exposed on surface of viruses similar to SARS-CoV that bind to human angiotensin-converting enzyme 2 (ACE2), which play a key role in the cell entry phase. In this study, we designed ACE2-derived peptides mimicking the binding site of ACE2 with S glycoprotein with affinity tags and linkers. The ACE2-derived peptides were produced in E. coli and purified. Their inhibitory effects were evaluated by performing neutralization assay and ELISA to compare the inhibitory effects. The most effective ACE2-derived peptides have potential for the application of inhibitors. |
