Polydiacetylene(PDA) liposome has made by self-assembly using photopolymerization process for gene delivery. PDA has specific feature that it changes its color upon different pH, temperature and ultraviolet irradiation. We have used 10,12-pentacosadiynoic acid(PCDA) derivative of PDA, coupling with polyethyleneimine(PEI) on the surface. PCDA and PEI were linked by 1-ethyl-3-[3-(dimethylamino)propyl] carbodiimide hydrochloride(EDC) and N-hydroxysuccinimide(NHS) coupling reagent. Agarose gel retardation and picogreen reagent assay were performed to make sure of cationic liposome and plasmid DNA complexation. Transfection efficiency study was carried out by protein assay and luciferase assay in HeLa and HEK293 cells. WST assay and confocal imaging were confirmed for cell viability and cellular uptake respectively in same cell line. From above studies, it is proven that our polymer vector will be a promising candidate for gene delivery. |
