Chemical derivatization with Girard's reagent T for the quantitation of gut microbial short-chain fatty acids by LC-MS/MS |

Short chain fatty acids (SCFAs) are the end products of fermentation by anaerobic gut microbiota, which are used as beneficial metabolites in regulating host physiological processes. However, SCFAs are difficult to analyze in mass spectrometric technologies due to their thermo-labile property and low molecular weights. Here, we have developed a sensitive and reliable method to quantify SCFAs by LC-MS/MS in the multiple reaction monitoring (MRM) mode. The SCFAs were chemically derivatized by the Girard’s reagent T (GT), which can provide a permanent cationic charge. The technique showed an excellent quantitative capacity, as determined by good linearity and limit of quantification for five SCFAs. The highly sensitive method would be primarily useful and applicable in quantifying SCFAs extracted from stool in an aqueous solution to monitor gut health. |
