Kinetic Separation in Escherichia coli by Developing Non-native Enzyme for Itaconate Production |
초록 |
Recently, microbial production of itaconatein Escherichia coli has been studied due to thecomplicated fermentation process and shortage of genetic engineering toolsof A. terreus. Unlike spatially compartmentalized itaconate synthesisof A. terreus, poor availability of its precursor, cis-aconitatehas been the major issue. In this study, directed evolution ofprotein known to have substrate promiscuity to citrate was conductedin E. coli to enhance the cis-aconitateavailability, with itaconate responsive screening system. Thereafter, themutant library was rationally designed and enriched. Finally,the developed mutant showed significantly enhanced catalytic efficiency tocitrate compared to the wild-type enzyme and consequently, its use enabledsignificantly increased itaconate production compared to the parental strain,indicating the successful kinetic separation between TCA cycle and itaconatebiosynthesis with the increase cis-aconitate supply for theitaconate production. |
