제목 hMSC-based immunomodulatorfor cancer therapy​
초록 Cytokine-basedimmune system activation requires high doses of cytokine to retain an appropriatelevel in vivo, thereby side effects such as cytokine storm may occur. Toovercome this limitation, we developed reactive oxygen species (ROS) produciblehuman mesenchymal stem cells (hMSC) for targeting tumor and locally secreting cytokine.We labeled the stem cell surface with polymer and photosensitizer (PPS) throughclick chemistry (PPS-hMSC). PPS-hMSC migrated to the tumor site by homing effectof hMSC, and pro-inflammatory cytokines such as heat shock protein 70 (Hsp70), IL-6,IL-8 are released from hMSC damaged by ROS under laser irradiation. These cytokinesinduce production and maturation of immune effector cells (IEC) such as T cell,natural killer cell (NK cell), B cell and antigen presenting cell (APC). Taken together,PPS-hMSC can be a potential agent being able to promote anti-tumor responses andtumor growth inhibitor for local immunotherapy.
