Effects of topography on osteogenic differentiation of mesenchymal stem cells and in vivo bone regeneration on chemically functionalized nanofibers. |
초록 |
In this study, we prepared BMP-2 immobilized electrospun poly (L-lactic acid) (PLLA) fibers with random and aligned structure to investigate the effects of chemical cues or their combined effects with topography on osteogenic differentiation of hMSCs and invivo bone formation. Florescent images showed that cells were adhered according to the underlying substrate topography in both BMP2 immobilized and non-immobilized groups. ALP activity and calcium content was higher on aligned fibers without BMP-2, however, BMP-2 immobilized groups showed no difference. Similar bone forming area was observed in implanted fibers which were dissected after two months of implantation in to an invio model. SEM and TEM images revealed that direction of regenerated bone and collagen fibers was followed the fiber orientation. Thus, we believe that, topography can direct cell adhesion and ECM deposition and chemical signals may be more prominent on cell function when there is a combination of these two. |
