제목 Functional characterization of glycosyltransferase (kanF) gene in kanamycin gene cluster of Streptomyces kanamyceticus
초록 The kanF (1164 nucleotides), located in the biosynthetic gene cluster of kanamycin producer, Streptomyces kanamyceticus, has putative function of glycosyltransferase. Multiple sequence alignment of amino acids shows 62 % and 33 % identities with already characterized neoD and btrM of Streptomyces fradiae and Bacillus circulans respectively. For the functional characterization of this gene, we attempted to over express in Escherichia coli BL21 (DE3) and the soluble fraction of protein was purified and used to carry out in vitro enzyme assay for glycosylation of UDP-N-Acetylglucosamine with 2-deoxystreptamine.
