A Plug-in Repressor Library for Precise Regulation of Metabolic Flux in Escherichia coli |

In metabolic engineering, improved production of value-added chemicals requires metabolic flux optimization. Although advances in synthetic biology have achieved the exploitation of a number of genetic elements for precise flux control, their use requires expensive inducers, needs complex and time-consuming processes to design and optimize conditions. To overcome this issue, we devised a plug-in phlF repressor library for target-specific flux control, where its expression level was diversified using degenerate 5’ untranslated region (5’ UTR) sequences generated by the UTR Library Designer. A validated wide range of repressor library was applied to enhance the production of 3-hydroxypropionic acid (3-HP) and lycopene in Escherichia coli. Consequently, we successfully achieved carbon flux optimization for efficient production. The most optimized strains produced 1.08 g/L of 3-HP and 10.6 mg/L of lycopene, which were improved 5.5-fold and 1.9-fold, respectively, compared to the parental strains. These results indicate that the repressor library is a powerful tool for efficient production of value-added chemicals by fine-control of carbon flux. |
