Accuracy,simplicity, sensitivity, quantification and cost-effectiveness are the mostimportant criteria for a genotyping method for single-nucleotide polymorphisms(SNP). One method developed for SNP genotyping, ligase-based coupled capillaryelectrophoresis (CE) is considered for clinical diagnosis. In this study, we developed a robust multiplex genotyping method using ligasedetection reaction and high-resolution CE-based single-strand conformationpolymorphism (CE-SSCP). With this high-resolution CE-SSCP system, we were ableto use similar-sized probes, simplifying the design step and assay process. Andwe found that this method could accurately discriminate single base mismatchesand quantifying analysis of SNPs of tp53 gene used as targets for multiplexdetection using multi-color fluorescence labeled probes. |
