Optogenetics is an emerging field that combines optics and genetic engineering to control genetically targeted cells with light. When specific light is illuminated on opsin channels of target neuronal cells, we can control channel opening and blocking by changing light wavelength. In this work, optogenetic system with carbon dot nanoparticle was developed for the non-invasive treatment of nervous system disorders. The optogenetic system was consisted with carbon dots (Cdots), supramolecular hyaluronate (HA) hydrogels, and two-photon laser for the control of channelrhodopsin (ChR2). To deliver and maintain Cdots in target region for a long time, supramolecular hydrogels was prepared with cucurbit[6]uril conjugated HA (HA-CB[6]) and diaminohexane conjugated HA (HA-DAH) tethering Cdots. We will present in vivo two-photon laser systems exciting Cdots in supramolecular HA hydrogels for the optogenetic therapy. |
