Optimized Silk/PLGA Hybrid Films for Human Aortic Endothelial Cells Culture |

A new compliant scaffold suitable for small-diameter vascular grafts has been developed that promotes strong attachment of endothelial cells. Silk will be used as a model protein for scaffold fabrication in this study due to its widespread versatility. Therefore, we fabricated hybrid polymeric films containing PLGA and various concentration of silk. The biocompatibility of silk/PLGA films was demonstrated by ELISA using TNF-α. The proliferation of cells studies supported the utility of this biomaterial films on HAECs. The preservation of cell phenotype on the silk/PLGA films was confirmed by the presence of cell-specific markers using immunocytochemistry and RT-PCR. The results indicate a favorable interaction between vascular cells and silk/PLGA films. These data confirm the potential of this novel hybrid films in vascular tissue engineering.
